Study On Oxidative Damage Protective Effect Of Flavonoids From Juglans Mandshurica Seed Shell

In this study,flavonoids were extracted from the Juglans mandshurica seed shell,and the purity was improved using macroporous resin.The protective effect of flavonoids on oxidative damage from the seed shell were studied by in vitro and in vivo experiments.(1)Favonoids were extracted by ultrasound assistance and purified by macroporous resin.The scavenging capacity of Juglans mandshurica seed shell flavonoids on ABTS free radical,DPPH radical,hydroxyl radical and reducing iron power were assayed by spectrophotometry.The antioxidant capacity in vitro was studied.The results were as follows.The effect of elution concentration on flavonoids purity was showed that:30%>50%>crude extract>70%.The ability of removing ABTS free radical,DPPH radical and hydroxyl radical and reducing iron power of 30%elution were stronger than other gradient elutes.The antioxidant activity of the ethanol gradient elution was positively correlateed with the purity of flavonoids.Among them,the antioxidant capacity of 30%elution was the strongest and can be used for the follow-up in-depth study of antioxidants in vivo.(2)Establishing the model of radiation-induced oxidative damage,the activity of SOD,GSH-Px,CAT,MDA and GSH in serum and liver tissue were measured by spectrophotometry after were irradiated 24 h later.The results showed that:The weight of mice in each group increased steadily,and the difference among the groups was not significant.It showed that the general physiological state of mice were not significantly effected by flavonoids of Juglans mandshurica seed shell.Compared with the model group,the activities of SOD,GSH-Px,CAT and content of GSH in serum and liver tissue were increased,and the content of MDA was decreased in every dose group of flavonoids of Juglans mandshurica seed shell.Those showed that flavonoids of Juglans mandshurica seed shell had protective effect on the antioxidant system of oxidative damage mice.(3)Establishing the model of radiation-induced oxidative damage,the effcet of flavonoids of Juglans mandshurica seed shell on the peripheral blood,liver,spleen,kidney and other tissues in mice were reseached by means of peripheral blood cell count and morphological methods after irradiation 14 days later.The results showed that:The flavonoids of Juglans mandshurica seed shell increased the content of RBC and HGB,but had no significant effects on WBC content.It can not only improve spleen and thymus atrophy in oxidative damage mice,increase organ index,but also reduce necrosis and degeneration of liver tissue as well as lighten atrophy and deformities of kidney glomerulus,and reduce floccule in kidney tubules.And it can promote down regulation expression of Bax protein and up regulation expression of Bcl-2 protein in liver and spleen tissue of oxidative damage mice,can inhibit the apoptosis of the liver and spleen cells.Those showed that flavonoids of Juglans mandshurica seed shell can repair and protect mice against oxidative damage.
