Reactive oxygen species are the byproducts of normal metabolism of oxygen and play a key role in the physiological and pathological processes. So far, it’s still great scientific and practical challenge towards real-time monitoring the redox equilibrium in vivo.Compared with other methods, Flourescent probes are efficient tools for detecting biological molecules due to their high sensitivity, high selectivity and less invasive for imaging intracellular processes of living celluar systems when associated with microscopic imaging techniques. This thesis firstly reported several newly designed fluorescent probes and applied in the detection of enzymatic activities and living cell fluorescence imaging.This paper is based on the changes of fluorescent spectra that organic small molecular fluorescent probe respond selectively to reactive oxygen species. The probes were successfully applied to fluorescence imaging HCl O in living cells. This article wased mainly accomplished from two aspects as follow.(1)A newly near-infrared fluorescent probe based on heptamethine cyanine for detection of hypochlorous acidWe designed and developed a nevol near-infrared fluorescent probe based on heptamethine cyanine Cy-Nph S. To explore the spectrum of the probe performance.Probe could provide highly selective and sensitive response to hypochlorite. Cy-Nph S has been successfully applied for detection of hypochlorous acid in RAW264.7 and MPO enzyme assy.(2)Two fluorescent probes based on fluorescein and its application in living cellSimulating activity center of glutathione peroxidase(GPx), selenium cysteine,based on fluorescein and 2,7-dichloro fluorescein,We designed and developed two reversible fluorescent probe for monitoring hypochlorous acid. Under physiological conditions, we explored the spectrum of the two probes, a large fluorescent increment(about 200 times) before and after reaction with hypochlorous acid. We also examined the reactivity of two probes with various ROS. They didn’t respond other reactive oxygen species and mercaptan but hypochlorous acid. The probes both show a selective specificity, high sensitivity, rapid response, the advantages of reversible response. On the other hand, the probe has been successfully applied for hypochlorous acid fluorescence imaging in living cell by fluorescent confocal microscopy.
