Selaginella uncinata (Desv.) Spring, which belongs to Selaginella, mainly contains flavonoids (especially biflavonoids) and phenolic compounds. The biflavonoids from Selaginella uncinata (Desv.) Spring has been proved to have the good anti-anoxic effect. For exploring the extraction technology of total flavonoids, reflux extraction technology of total flavonoids and extraction of total flavonoids with cellulase about Selaginella uncinata(Desv.) Spring was investigated to get the optimal conditions. In the aspect of total flavonoids enreachment, extraction residure was dissolved in water and then portioned successively with petroleum ether, chloroform, EtOAc and n-BuOH. The total flavonoids sample (SU-E-F) was purified by Sephadex LH-20 column chromatography from the EtOAc part. The results showed that the flavonoids content of SU-E-F was higher, which was higher than the one of extraction residue. And the method basically met the objective of total flavonoids enrichment. This study which was mentioned above was taken as the scientific basic for further developing and industrial production of total flavonoids from Selaginella uncinata (Desv.) Spring.Meanwhile, extraction residure of Selaginella uncinata (Desv.) Spring, petroleum ether part (SU-P), chloroform part (SU-C), EtOAc part (SU-E), n-BuOH part (SU-B), water part (SU-W), total flavonoids sample (SU-E-F) by enrichment method and non-flavonoids sample (SU-E-N) were detected about the initial anti-caner activity. The results showed that the anti-caner activity of total flavonoids sample (SU-E-F) was better than others and indicated that the flavonoids from Selaginella uncinata (Desv.) Spring had good anti-cancer activity. This found was taken as the basic for further study on the anti-cancer activity of specific flavonoid from Selaginella uncinata (Desv.) Spring and also supllied the objective research material for further study between anti-anoxic effect and anti-cancer effect.And on this basic, the high polar constituents from Selaginella uncinata (Desv.) Spring, which was focused on rarely in the past, was studied. The SU-B was isolated and purified by the use of silica gel column chromatography, Sephadex LH-20 column chromatography, pre-HPLC and so on. The structures of isolated compounds were identified through spectral analysis and physical-chemical properties. Three compounds were isolated and identified. They were, respectively, (3β,12β, 25R)-spirost-5-ene-3,12-diol-3-O-α-L-rhamnopyranosyl-(1â†'2)-O-[α-L-rhamnopyran osyl-(1â†'4)]-O-β-D-glucopyranoside (Compound I), (2α,3β,12β, 25R)-spirost-5-ene-2,3,12-triol-3-O-α-L-rhamnopyranosyl-(1â†'>2)-O-[α-L-rhamnopyranosyl-(1â†'4)]-O-β-D-glucopyranoside (Compound â…¡) and (3β,7β,12β, 25R)-spirost-5-ene-3,7,12-triol-3-O-α-L-rhamnopyranosyl-(1â†'2)-O-[α-L-rhamnopyranosyl-(1â†'4)]-O-β-D-glucopyranoside (Compound â…¢). Compound Ⅱ and compound â…¢ were the new compounds; compound I are obtained from the genus for the first time. They were steroidal saponins, and Compound â…¡ and compound â…¢ were isomeric compounds. The study would benefit the knowledge of Selaginella uncinata (Desv.) Spring, supply the material base for activity of Selaginella uncinata (Desv.) Spring and be taken as the scientific base for further study on high polar constituents from Selaginella uncinata (Desv.) Spring.
