A large number of steroidal saponins were contained in the root of Smilax Scobinicaulis C.H.Wright,and among them 6-ketone steroidal saponins were rich in content,which had a carbonyl function at position C-6.This made 6-ketone steroidal saponins spirostanic analogue of the v teasterone.They had same functional groups with teasterone type of BR sterol compounds,and has a similar activity.Early research has shown that this kind of compounds had significant biological activity,for example promoting seed germination and seedling growth,etc.So it could be used as plant growth regulator.Besides this,the synthesis of Laxogenin and its glucoside was much easier than and its glucoside.So exploitation and utilization on 6-ketone steroidal saponins was of great significance.This article established 6-ketone steroidal saponins content determination method by screening the chromogenic agent,central composite design-response surface method to optimize chromogenic condition.The steroidal saponins extract soluble concentrate from the rhizome of Smilax Scobinicaulis C.H.Wright was studied through the screening of solvents and SAA,And then to determine efficacy of activity.The main test contents are as follows:1.Got the content determination method of 6-ketone steroidal saponins.Compared with the existing literature,made anisaldehyde-sulfuric acid ?(A reagent:0.5%anisaldehyde ethyl acetate solution,B reagents:50%sulfuric acid ethyl acetate solution)as the chromogenic agent for the test,filtrating by the visible light wavelength scanning,precision and stability test,and got the maximum absorption wavelength of 429 nm.The best color conditions wre got through single factor experiment and chromogenic condition optimization:dry up solvent of samples,add 2 mL ethyl acetate,shake well,in order to join 1.2 mL of A and B reagents,69.5 ? water bath react for 26.3 min,then took into the ice water bath for 10 min,to make the reaction stop,the chromogenic system was determined at 429 nm absorbance value,and calculated the content.2.Determined the formula for 0.1%soluble concentrate from 6-ketone steroidal saponins.Used the solubility of effective ingredients in the solution to screen the solvent type and proportion.Determined the variety and dosage of emulsifier by the low temperature stability,heat storage stability and dilution stability.And then determined the composition of the soluble liquid agent:saponins extract in 0.5%(specific volume),?-cyclodextrin(molar ratio of 1:1 with saponins)in 0.64%,polysorbate-80 15%,anhydrous ethanol 30%,and dimethyl sulfoxide up to 100%.After the experiment proved that the various physical and chemical indexes of soluble concentrate accorded with the relevant national standards.And the standard of quality analysis for soluble concentrate was prepared.3.Tested biological activity of 0.1%soluble concentrate from 6-ketone steroidal saponins.Due to the suitable concentration from 6-ketone steroidal saponins could promote radish seedling growth,the increments of radish seedling cotyledon,hypocotyl length and diameter were studied to estimate the effects of different concentrations of aqueous solution and determine the optimal concentration.The results showd that the 10-7 mol·L-1 soluble concentrate(The medicament was diluted 104 times)was the best,and if the concentration increase or reduce,its effectiveness would be influenced.
