Since early 1980 s,researchs began to drown more and more attentions to functional carbohydrate that has special physiological functions.The thesis explored the effect of jujube dietary fiber on intestinal flora and functional constipation and on immune function of jujube oligosaccharide in animal experiment.Method:1.The effect on intestinal flora of jujube dietary fiber was investigated according to the following methods.Mice were randomly divided into 4 groups including control group and jujube dietary fiber groups at the low,middle and high doses.The dosage of jujube dietary fiber were 0.30,0.60 and 1.20 g per kilograms of body weight for low,middle and high dose groups respectively.The jujube dietary fiber was given for consecutive 14 days,while the control group mice were given same volume of distilled water instead of jujube dietary fiber.The fresh feces of mice were colleced on the 0th day and 14 th day of gavage to homogenized and diluted,then inoculated to have the bacteria growed and indentified.The number of intestinal flora,such as Bifidobacteria,Lactobacillus, Enterobacter,Enterococcus,and C.perfringens in each fecal sample was quantified.2.The effect on functional constipation of jujube dietary fiber was investigated according to the following methods.Mice were randomly divided into 5 groups including control groupconstipation model group and jujube dietary fiber groups at the low,middle and high doses.The dosage of jujube dietary fiber were 0.30,0.60 and 1.20 g per kilograms of body weight for low,middle and high dose groups respectively.The jujube dietary fiber was given for consecutive 10 days,while the control group and constipation model group mice were given same volume of distilled water instead of jujube dietary fiber.Functional constipation model was established after oral administration of jujube dietary fiber for 10 days.The intestinal motion rate,the first defecation time and feces weight of all the mice were investigated to evaluate the effect of functional constipation of jujube dietary fiber.3.The effect on immune function of jujube oligosaccharide was investigated according to the following methods.Mice were randomly divided into 4 groups including control group and jujube oligosaccharide groups at the low,middle and high doses.The dosage of jujube oligosaccharide were 0.50,1.00 and 2.00 g per kilograms of body weight for low,middle,and high dose groups respectively.The jujube oligosaccharide was given for consecutive 28 days,while the control group mice were given same volume of distilled water instead of jujube oligosaccharide.After 28 days,the effect on immune function of jujube oligosaccharide was evaluated from 3 aspects of humoral immunity,cellular immunity and phagocytic function of macrophages.Results:1.Jujube dietary fiber’s effect on intestinal flora.Jujube dietary can reduce contents of Enterobacteriaceae,Enterococci and C.perfringens and promote the proliferation of Bifidobacterium and Lactobacillus at the administration of 0.60 g/(kg·bw).Compared to control group,the contents of Enterobacteriaceae in low,middle and high dose groups were reduced by 7.96%,15.04% and 12.83%; the contents of Enterococci were reduced by 3.53%,4.37% and 6.86%;the contents of Clostridium were reduced by 2.82%,5.63% and 7.57%.At the same time,compared to control group,the contents of Bifidobacterium in low,middle and high dose groups were improved by 1.40%,2.79% and 3.37% and the c o n t e n t s o f L a c t o b a c i l l u s w e r e i m p r o v e d b y 1. 0 2 %, 2. 4 2 % a n d 3. 3 2 % 。2.Jujube dietary fiber’s effect on functional constipation.The motional rate of ink in the intestine was enhanced by jujube dietary fiber administration,initial defecation time was abridged,the quantity of fecaland total,dry feces weight increased when compared to the model control group.The intestinal motion rate in low,middle and high dose groups were increased by 14.96%,36.74% and 46.61%.The initial defecation time were abridged by 10.72%,22.37% and 19.24%.The total feces weight in 6 hours were increased by 41.86%,65.12% and 111.63%. The dry feces weight in 6 hours were increased by 21.88%,56.00% and 92.00%.The quantity of fecal in 6 hours were increased by 37.10%, 79.38% and 77.37%.3.Jujube oligosaccharide’s effect on immune function.High dose(2.00 g/(kg·bw))of jujube oligosaccharide can significantly increase plaque forming cell of spleen and phagocytic index by 5.02% and 33.72%.It also can highly significantly improve the antibody level,the thickness of footput and spleen lymphocyte transformation function in mice by 26.51%,50.70% and 58.33%.Middle dose(1.00 g/(kg·bw)) of jujube oligosaccharide can significantly increase spleen lymphocyte transformation function by 16.67%.It also can highly significantly improve the antibody level and plaque forming cell of spleen by 11.60% and 24.33%.Low dose(0.50 g/(kg·bw))of jujube oligosaccharide can significantly increase the antibody level in mice by 10.38%.Conclusion:The administration of 0.60 g/(kg·bw) of jujube dietary fiber is beneficial for adjusting intestinal flora and relieving functional constipation.The administration of 0.50 g/(kg·bw) of jujube dietary oligosaccharide can improve immune function of mice.
