Sporisorium reilianum is a fungus.It can provide nutritional elements for human body as a natural health food,and has various physiological functions such as antitumor,anti-inflammation,hypolipidemic,and hematopoietic enhancement.In this study,Sporisorium reilianum was used as raw material,and the active ingredients were analyzed and polysaccharides were extracted.The antioxidant activity of polysaccharides,which was separated and purified by ion-exchange chromatography,was determined.The dietary fiber of Sporisorium reilianum was prepared,and the physicochemical and in vitro functional properties were determined.Through in vivo experiments,the regulation of Sporisorium reilianum on intestinal health was studied.(1)The active ingredients and ABTS free radical scavenging were analyzed,using water,70%ethanol and methanol as the extraction solvent,and the content of total phenol and polysaccharides as indexes.The purified Sporisorium reilianum polysaccharide(SRPS)was obtained by water extraction and alcohol precipitation.After separating and purifying by ion-exchange chromatography,the purity of the polysaccharides was determined by spectrophotometry and the polysaccharides were qualitatively analyzed by infrared spectroscopy.The results showed that:The content of polysaccharides in water extract was the highest and ABTS free radical scavenging was the strongest.After further ion exchange column chromatography,four polysaccharide components of SRPS1,SRPS2,SRPS3 and SRPS4 were isolated and all had pyranose structure.The polysaccharides content of SRPS1 eluted with deionized water was the highest,indicating that the Sporisorium reilianum polysaccharide was mainly neutral polysaccharides.Meanwhile SRPS1 had the highest purity,which was 9.86%higher than that of refined Sporisorium reilianum polysaccharides.(2)The antioxidant activity of Sporisorium reilianum polysaccharides in vitro was evaluated by measuring the ABTS free radical,nitrite scavenging effect,total reducing power and copper ion reducing power.The results showed that:The antioxidant capacity of Sporisorium reilianum polysaccharide increased with the increase of the mass concentration,and the scavenging effect on nitrite was stronger than that of BHT.The ability to reduce copper ion was greater than that of VC and BHT when the concentration was higher than 27 mg/mL,indicating that Sporisorium reilianum polysaccharide had a significant antioxidant effect.(3)The soluble and insoluble Sporisorium reilianum dietary fiber was prepared by enzyme-weight method.The physicochemical properties of Sporisorium reilianum dietary fiber were studied by measuring the swelling power,water holding capacity and oil holding capacity.The in vitro functional properties of Sporisorium reilianum dietary fiber were studied by measuring glucose adsorption capacity,cholesterol adsorption capacity,sodium cholate adsorption capacity and glucose dialysis delay index.The results showed that:The swelling power and water holding capacity of soluble dietary fiber in Sporisorium reilianum were significantly higher than that of Pleurotus eryngii,and the glucose adsorption capacity and dialysate delay index were higher than that of Pleurotus eryngii.The swelling power of insoluble dietary fiber in Sporisorium reilianum was significantly higher than that of Pleurotus eryngii,and the glucose adsorption capacity was higher than that of Pleurotus eryngii,indicating that Sporisorium reilianum dietary fiber had better physicochemical and in vitro funcitional properties.(4)The regulation effect of Sporisorium reilianum polysaccharide on intestinal health in normal mice was evaluated by measuring mouse feces pH,water content and observing colon tissue morphology.Dextran sodium sulfate(DSS)was used to establish the model of inflammatory injury in mice.The regulation effect of Sporisorium reilianum on intestinal health in inflammatory injured mice was evaluated by evaluating and measuring the activity of superoxide dismutase(T-SOD)and myeloperosidase(MPO)in colon tissue,observing the changes in colon tissue and measuring the intestinal flora in mice through macro indicators.The results showed that:The Sporisorium reilianum polysaccharide could properly regulate the body weight of normal mice,reduce fecal pH and increase fecal moisture content.Sporisorium reilianum could reduce the macro index scores in mice with intestinal inflammatory injury,increase the activity of SOD in serum and colon,and reduce the activity of MPO.It could prevent colonic tissue damage in mice with intestinal inflammatory injury and significantly increase the intestinal flora richness and diversity.It showed that Sporisorium reilianum had the functions of preventing intestinal inflammatory injury,protecting intestinal tissue,regulating intestinal flora and promoting intestinal health.
