Antifungal proteins were extracted from seeds of naked oat by water extraction method. In order to make sure the optimal water extraction conditions, the single factor experiment and response surface analysis were used by extraction time, extraction temperature, pH, liquid-to-solid ratio. The purification of antifungal protein of seeds of naked oat was done by cation exchange chromatography. Main results were as follows: (1)Response surface analysis was used to get the optimal water extraction conditions. The optimal water extraction conditions are:the extraction temperature is 30 ℃;pH is 7;liquid-to-solid ratio is 1:12;extraction time is 12h.The diameter of inhibition zone was 18.10 mm in this condition.(2) The separation and purification of antifungal protein of seeds of naked oat was done by cation exchange chromatography. The results showed that 3 peaks were acquired by gel filtration, only I peak had inhibition activity with inhibition zone diameter 25.12 mm. IC50 was 0.2385mg/mL. naked oat antifungal protein molecular weight was 37KDa by SDS-PAGE.(3) The purification of naked oat antifungal protein was used by the stability testing. Result:The high temperature(>40℃)and acid environment had a significant effect on inhibition activity. And Nacl concentrations had no effect on inhibition activity.
