At present, the use of computer detection instead of artificial had formed a trend in the field of glutamic acid fermentation is also seems particularly outstanding, the computer control of the glutamic acid fermentation is hot.But at this stage is not high temperature resistant of biological sensors to provide the testing results of biomass, nor is stable and reliable testing instruments to real-time quantitative biochemical indexes and state variables of glutamic acid fermentation, the fermentation encountered bottleneck of computer control. At present, enzyme electrode method on-line detection of biomass technology was applied to glutamic acid rods coli (Corynebacterium gulutamicum) S9114 glutamic acid fermentation is rare.According to the situation of described above, this study selects the SBA-66 series biomass on-line detection system to online real-time detection of glutamic acid fermentation, SBA-66 series biomass on-line detection system for the shandong academy of sciences institute for biological studies on its own research and development production.On-line detection and the result of the fermentation process of pH, dissolved oxygen, temperature change of other parameters, such as correlation, mutual guidance amino acid fermentation, provide a basis for the practical data in glutamate fermentation optimization.The task of this study and the results are as follows:1. This study use the high yield glutamic acid after mutagenesis, glutamic acid fermentation strains, combined with the SBA-66 series biomass detected the OD value of online detection system, detection of lactic acid content, content of glutamic acid and glucose content in the fermentation process, detection in real time, the sampling results at the end of the measurement automatically saved.Set the SBA-66 series of the parameters of biomass on-line detection system for T= 66 s, sampling time T sample interval-66 min, deliver time T= 66 s, T= 1 s sampling time, T= 30 s emptying time.Switch system is adopted, electromagnetic valve, to control the sample liquid flow is driven by peristaltic pump, and the use of the system the human liberation, reduces the industrialized production of artificial cost;In addition, the system of the measurement cycle can be set up, the result is accurate, the result of the on-line detection and the fermentation process of pH, dissolved oxygen, temperature, the change of other parameters, such as correlation, mutual guidance amino acid fermentation, provide a basis for the practical data in glutamate fermentation optimization.
