Guided by the pancreatic lipase inhibitor activity screening, black tea collected from different factory in Guizhou province, was fractioned by solvent partition, different chromatography methods.Firstly, methanol extract of black tea was partitioned by petroleum ether, chloroform, ethyl acetate and water, and resulted in five fractions, and their inhibitory on pancreatic lipase were detected. It was found that the ethyl acetate fraction showed the most potent activity, and it was determined as the active fraction of black tea extract. With different chromatography methods, the active ethyl acetate fraction was separated into active compound caffeine and several active subfractions according to the pancreatic lipase inhibitory screening results. The content of polyphenol and caffeine were measured by by Folin phenol method and UPLC method. The relationship between pancreatic lipase inhibition and constituents of the fractions were discussed. In order to described the pancreatic lipase inhibition activity, the inhibition mode of the active ethyl acetate fraction and the typical components of black tea, caffeine, EGCG and theaflavin evaluated.The results showed that: ①ethyl acetate fraction is the active fraction of black tea of inhibitory against pancreatic lipase.②Caffeine is the main active compound in the black tea, and the contents of polyphenol and caffeine in the black tea extracts are correlated with the pancreatic lipase inhibitory, the inhibition activity was increased as the content of the polyphenol and caffeine increased; ?The inhibition modes of ethyl acetate fraction and its mainly constituents, caffeine, EGCG and theaflavin are all uncompetitive.
