Chitosan is a kind of polycation straight-chain polymer,the reserves are abundant with excellent physical and chemical properties in nature.The loaded-drug of chitoan materials exist broad application prospects,But the drug-loading chitosan materials are limited by its poor water soluble in biological mdicine materials application,the biocompatibility and biological toxicity of modified chitosan has been the topic of research and attention.In this study,we used a novel pinhole extrusion coagulation precipitation to prepare CS/CMC composite gel microsphere were characterized by FTIR,XRD,SRM.we discussed the different factors on the morphology and size of composite gel microphere.Through the BSA as the drug,studied the encapsulation efficiency and load efficiency of BSA-loaded CS/CMC composite gel microphere and the release behavior in the different releasing medium.we prepared the Largazole-loaded doping TPP CS microsphere by ionic gel mehod,studied the release behavior of Largazole in the phosphate buffer solution and the animal activity in mice.Through the analysis test of FTIR,XRD,SEM,the study showed that the CS/CMC composite gel microsphere were successfully prepared.This study explored the titration height,extrusion speed,coagulating liquid flow pattern,chitosan concentration,pinhole diameter,coagulating liquid concentration on the influence of the CS/CMC composite gel microsphere,used the orthogonal experiment to optimize the condition.The optimum conditions were:titration height was 10 cm,extrusion speed was 2s/drop,the flow pattern of coagulating liquid was static when extrusion and the opposite was moving,chitosan concentration was 2(w/v%),the pinhole diameter was 0.33 mm,the coagulation liquid concentration was 0.5(w/v%).Through the orthogonal experiment analysis,showed that the greatest impact on the size was chitosan coagulation liquid concentration,followed by chitosan concentration and pinhole diameter.This studied that the encapsulation efficiency of BSA-loaded CS/CMC composite gel microsphere reached 78.8%.The releasing behavior of the physiological saline and phosphate buffer solution showed that it existed excellent controlled-release,and the accumulative releasing rate was more faster in phosphate buffer,the CS/CMC composite gel microsphere has less resitance to the nature of the physiological saline.This study prepared the Largazole-loaded doping TPP chitosan microsphere by ionic gel method,the encapsulation efficiency reached 95.67%.When the microsphere was released in simulating intestinal phosphate buffer solution,the accumulative releasing rate reached 84.57%,the releasing time reached 168 h.The assessment of animal experiment showed that Largazole-loaded doping TPP chitosan microsphere presented a good therapeutic effect,the Largazole had excellent biological activity over 7-day period...
