Chitosan is a product of part of chitin and a kind of high content,low cost of polysaccharide.It widely exists in the nature and it is the second largest renewable resource on earth after cellulose.It is also now found that the only natural basic polysaccharide.It has good biocompatibility,biodegradability,hemostatic properties,antimicrobial properties,adsorption,avirulent insipidity,.relatively stable physical and chemical properties.It is widely used in various fields such as biology,medicine,chemical,food,cosmetics.This paper mainly studies the coagulant energy of chitosan derivatives,dicuss the possibility of using it as a new type of textile hemostatic dressing.N-alkyl chitosan with different carbon chain length alkyl was prepared by reductive amine method.Adapting infrared spectroscopy(FTIR),nuclear magnetic 1H NMR(1H NMR)and elemental analysis to perform structural analysis and characterization of materials in order to prove that alkyl chain has been successfully grafted to the main chain of the chitosan and the conclusion that substitution degree increases with the rising of mole ratio of alkyl aldehyde and chitosan amino.The cytotoxicity was measured by MTT colorimetry and cell fluorescence staining.It was initially evaluated whether it could be used as a biohemostatic material.The results showed that the prepared n-alkylation chitosan was not toxic in this paper,and it can be used for medical textile dressing.To study the coagulation performance of N-alkyl ated chitosan,methods of whole blood coagulation time,thrombus elastic diagram(thromboela-stogram,TEG)and rheological test were used to carry out comparative analysis of different materials,It was found that N-alkylated chitosan is better than chitosan in coagulant effect and the fact that the greater the degree of substitution and the longer the alkyl chain grafting,the better the coagulation effect and the shorter the time shrinkage fibrinogen.The coagulation mechanism of the material was detected by testing APTT(activated partial boplastin time),PT(prothrombintime),TT(thrombin time),and intracellular Ca2+ concentration in platelet cells and the expression of P selection in the membrane.The results showed that the coagulation effect was not dependent on the traditional internal and external coagulation pathways,which would not cause significant changes in the expression of the concentration of Ca2+ concentration in platelet cells and P selection based on the surface above platelet membrane.
