| The distribution and technology of extraction and antitumorous in vitro of Iimonoids in citrus were investigated. The results showed that: 1. The content of limonoids was in seed higher than that of in pericarp and that of in juice was lowest in citrus. The turns of content of liomonoids in varieties as follows: Maxu grapcfruit>Guanxi pummclo> Nancbong sweet navel> Jincheng> Tangerine orange. 2. The extraction limonoids were identified as limonin (fraction I ) and nomilin (fraction II) byTLCandlRand NMR. 3. Extraction and purification processing of limonoids in citrus seeds: Dry seeds ground--efat with petroleum exfracted with acetone concentrate to dry at depression defat with petroleum filter dissolve and crystallization with CH2Cl2/2-propanol step ciystallization separate with CH2C12/2-propanol mother liquter heating and solvent with 2-propanol ,filter fLC pure limonin I,COO1 and crystallize stcp crystallization with methanol separate with TLC pure nomilin 4. Through orthodoxy experiment, the best condition of defat is : temperature at 25C, pH7.0, defat 10 hours. Through combination rotational design of experiment, the best technology of extraction as follows:(1)limonin: temperature at 35 C , extraction 25 hours, acetone 47 mug. (2)nomilin: temperature at 30 C , extraction 30 hours, acetone 47 mug, the best technology of extraction limonoids as follows: temperature at 30C, extraction 30 hours, acetone 47 mug, At these conditions, the production was 370.75-372.66 (mg/kg) 5. The effects of limonin and nomilin on antitumor in vitro were significantly inhibition on human hepatic carcinoma cell line and human bladder carcinoma cell line and the nomilin was the best. The effect on inhibition increased with the content of limonoids and cell culture time. The effect on human hepatic carcinoma cell line was better than human bladder carcinoma cell line. The inhibition effects of limonin and nomilin on fibroblast cell line were not significant. |
