Study Of The Purification Process For The Extraction Of Egg Yolk Lecithin And Lecithin

Phosphatidylcholine, as nature emulsifying agence and nourishment,is a sort of phospholipids. It is affluent in brain, heart and marrow ofpure animals, the barm and the yelk of birds egg. The nature high -pWied-phosphatidylcholine is used in food industry, pharmacy and so on. At present, almostly all of the high-pure phosphatidylcholine used in our country are imported, the annual demand is upward to several tons, so preparing high-pure phosphatidylcholine is our main purpose.With egg yelk as raw material, extracted by aquiferous ethanol and aether, removing water and oil by acetone, we gain phospholipids mixture, and search the extracting conditions. The optimum condition is determined:extracting temperature (20 ), time (60mins), solvent (92%ethanol),solvent quantity (12 times of the yelk), at which yield is highest; using thinlayer chromatography to measure its component, the content of phosphatidylcholine extend to 68.2%. Then using phospholipids containing 25% lecithin as raw materials, the solvent mixture(ch,proform/methanol) as eluate, using chromatography column on silicic acid, we search the optimum conditions of gain high-pure lecithin. At ground of the condition, we separate the phospholipids prepared by our method, after evaporated by freeze-dry machine, ultimately gain high-pure white lecithin, the final yield is 89.47%. Analysised by thin layer chromatography, there is only one spot, using thin-layer chromatography-scanning method to measure it, the purity is higher than 97.4%.The process of extraction is reasonable, the cost is low, and the content of lecithin in phospholipids extracted is higher than 60%. And at the laboratory, using chromatography column on silicic acid to purifc% lecithin, the purity and final yield of the product are higher, operation issimple, and the raw material can be gained easily. These establish the foundation for industrialization.
