Hydrogenated phosphatidylcholine (HPC) is widely used , mostly as feed and food additives, also as carrier of ribose nucleoprotein. It is also a very important raw material in producing transfusion. In this paper, we studied the preparation and purification technology of high purity hydrogenated soybean phosphatidylcholine.The 70.5% soybean phosphatidylcholine was used as raw material which was catalytically hydrogenated. Solvent, temperature, pressure, catalyst and reaction time were examined. The optimal conditions were as follows: 10 grams of soybean phosphatidylcholine with an iodine value of 85 was dissolved in 160 ml of mixed solvent of dichloromethane and ethanol (3:1, v/v) and agitated under a hydrogen pressure 0.5 MPa, temperature 50 ± 2 ℃, in the presence of 5% palladium on charcoal for 3.5 h, a solid product with an iodine value below 10 and PC purity of 65% was obtained.To obtain high purity HPC, we used column chromatography to purify HPC. Silica column chromatography was selected as a primary method for the purification of soybean HPC. At this step, mixed solvent of petroleum ether (60~90℃) and 2-propanol (1/1 ,V/V) was used as the mobile phase. Thus, when HPC/silica (w/w) was 0.064, sample concentration was 40 mg/ml, the product purity was over 80%, and the recovery of HPC was nearly 70%.High purity HPC was prepared by alumina column chromatography. We selected the chloroform-methanol (9/1 and 1/9,7/3 and 3/7) as mobile phase. When flow rate was 1.5 ml/min, HPC/alumina was 0.03 (w/w), the product purity was about 90%, and the recovery was 60%.
