A water soluble crude polysaccharide FA has been isolated from the caput-medusae of Ganoderma tsugae Murr. The crude polysaccharide has been fractionated by smelting after freezing and ethanol, so FA2 was abtained. FA2 was deamylumized and examined by gel filtration on Sephadex G-100. The results indicated that FA2 was homogenous. FA2 has been purified by gel filtration on Sep G-100G. C. analysis indicated that FA2 was composed of Gal and Glc in the molar ratio of 8.09:1.00. FA2 was further studied. In the structure analysis of FA2, hydrolysis with hydrolysis with amylase and hydrolysis with fibrin enzyme, partial hydrolysis with acid, periodate oxidation, Smith degradation, I.R, G. C. , methylation, and G. C.-MS analysis were the principal methods used.The results revealed FA2 is a kind of high branched structure. The main chain is made up of Gal and Glc, the inner is (1--3) -linked-Gal, the border is (1--6)-linked-Gal , (1--6)-linked-Gal and (1--3)-linked-Glc residues are substituted at 6-0. On average, there is 4 branch among 10 main chain residues. The side chain is composed of (1--6)-linked-Gal, (1--3, 6)-linked-Gal, ( 1--3 ) -linked-Glc, (1-3,6)-linked-Glc. The nonreduced end is composed of Gal. |
