Solanesol is an important intermediate material of medicine. It has been isolated from flue-cured tobacco primarily and has a good applied prospect.The thesis presents a study on extraction and analysis of solanesol based on the demands of enterprise. The creative works are summarized five aspects as follows:1. We developed a method for the determination of solanesol based on Capillary Gas Chromatography. By using temperature program and selecting PEG-20M column the sample was well separated . This method is fast, accurate and reproducible. It has good linear relationship in the range of 0.2-2ug . The result has shown that the relative standard deviation was 1.73%.2. Select a new solvent for extraction. The orthogonal design of four factors and three levels were used to select the optimal extraction method. The best extraction process is : the ratio of v/m is 13; the extraction time is 10h; temperature is 50; the radium of tobacco particle is 68.75um. The method is stable and easy to operate.3. A widely applied kinetic equation for Chinese traditional medicine extraction process was used in solanesol extraction from tobacco leaves firstly. Experiment data could perfectly match with this kinetic equation, the rate-determining step for extraction solanesol from tobacco leaves was the diffusion of solanesol through the tabacco particle . The apparent activation energy for solanesol diffusion of process was 18.64kj/mol .So it presents a theory basis for extraction process.4. A new purification process of extracted solanesol has been developed in this work. Mainly four steps are contained in this procedure, i.e saponification, extraction, wax removing and recrystalization, the progress was studied systematically first time. The optimal condition was determined and the cheapest solvent was optimized. Indicating that solanesol does not change during the process but the content ofit rises greatly.5. Optimized the operational condition of HPLC and TLC. By selecting the ultra-violet detector, the time for stablization of instrument was shortened; by studying on the developer and developing temperature, the optimal thin layer condition was determined. It's rapid, reproducible.
