The tobacco industry has made every effort to reduce the amount of tar in flue gas through various technical means,resulting in the problem of insufficient aromas.An important prerequisite for flavoring and adding fragrance to tobacco leaves is a full understanding of the composition and content of aroma components in tobacco.This paper takes Yunnan Baoshan Honghua Da Jinyuan tobacco leaf as the research object.First of all,the pretreatment methods of aroma components in tobacco leaves were investigated in order to obtain more aroma components of tobacco leaves.Secondly,in order to solve the problem of limited capacity of the gas chromatogram single column chromatography,a method consist of supercritical fluid chromatography and gas chromatogram/mass spectrometry(SFC-GC/MS)was developed.And this method was used to analyze the composition and content of aroma components of Yunnan typical tobacco.Monomeric compounds of representative tobacco leaves was separated by phytochemical methods and high performance liquid chromatography.Providing technical support for cloud-produced cigarettes.The main research contents as follow:(1)Experimental conditions of soxhlet extraction(SE),simultaneous distillation extraction(SDE)and ultrasonic extraction(UE)were optimized.The differences of the compositions and contents of the aroma of tobacco by these three methods were studied.There were 80 kinds of compound were found by SE,58 kinds of compound were detected by SDE and 59 kinds of compound were detected by UE.The total content of compound was 4755.97 ?g/g by SE.And the total content of compound was 721.85 ?g/g by SDE,while the total content of compound was 284.04?g/g by UE.Therefore,SE method was chose for subsequent experiments.(2)The working conditions of the SFC were optimized.UV-Vis detector with a wavelength of 210 nm.9% methanol was used as the modifier.The total flow rate was 30 m L/min and the back pressure was 100 bar.The aroma components of tobacco was extracted by SE.A part of extraction was detected by GC/MS.Another part of extraction was divided into 5 sections by supercritical fluid chromatography(SFC)and every section was detected by GC/MS(SFC-GC/MS).The results showedthat there were 71 kinds of aroma components was detected by GC/MS,while 130 kinds of aroma components was analyzed by SFC-GC/MS.(3)The composition and content of aroma in different varieties of Yunnan tobacco were analyzed by SFC-GC/MS.There are some differences between different tobaccos of Yunnan.This work provides a technical reference for cigarettes.(4)Three new compounds and nine known compounds were isolated from tobacco by liquid chromatography and supercritical fluid chromatography.We found thecompound2-(4-hydroxy-3,5-dimethoxyphenyl)-3-methyl-5-(3-prenyl)benzofuran-7-olhasanti-tobacco.The research enriched the chemical constituents of tobacco and provided a reference for the scientific research of tobacco.
