S-adenosyl-L-methionine(SAM), an important metabolic intermediate in the metabolic flux of sulphur is involved in many metabolic pathways .An recombinant strain which can produces high level of SAM was obtained. The fermentation conditions that can effect the microorganism growth and the SAM yield was studied. The experiment on purification of SAM with macroporous resin was completed.The culture conditions of the recombinant Pichia pastoris in fermentor (work volume 5L) was optimized in terms of pH, dissolved oxygen, Glycerol and methionine in fermentation broth and the SAM production of the recombinant cells reached 4.3 g/L after fermentation for three days under the optimized conditions.During the purification process with column chromatography, the different types of macroprous ion-exchange resins were compared and then the best (D151) was chosen out, as well as the mechanism of exchange was discussed .The operation parameters on absorption and elution were established. A whole production routine including extraction, ion exchanging chromatography, two-step elution, and precipitation was put forward .The purity of the refining product can be attained to 91.0%.
