Numerous free radicals are generated in the course of biology metabolism. Superoxide anion radicals (02" '). hydroxyl radicals (HO), hydrogen peroxide (H2O2) and ROO are four typical ones. They have important relation with aging and many diseases. It is well known that reactive oxygen species have damages on organisms and have gotten great attention in the 20th century, but the short lives and low concentration of free radicals mean to the determination difficult, especially the determination in vivo and in situ has no breakthrough. In addition, there exist the anti-oxidation systems preventing the damages of free radicals in organisms. Up to date, it cannot be observed the dynamic generation of reactive oxygen species in some organs. Enough cognition and sufficient experimental evidences of their generation mechanism, physiological action and dynamic damages on organisms are not acquired. Therefore, the therapy effects of many diseases in clinic are limited and they are main obstacles to the plant cultivation in adversity. So it is one of challenging scientific researches in analytical chemistry, in which novel analysis and determination technologies contraposing to reactive oxygen species in vivo need to be developed.Based on the changes in spectrum characters of the fluorescent probes reacting with reactive oxygen species, we have carried out three aspects of investigation:First, two fluorescent probes of O2- vanillin-8-aminoquinoline and 2-(2-pyridil)-benzothiazoline were synthesized. Their reactions with O2- ' were investigated by spectrofluorimetry and flow injection spectrofluorimetry. The reaction mechanism was studied and the proposed methods were applied to the determination of SOD activity.Second, baded on sodium terephthalate trapping HO, an automated spectrofluorimetry for determining HO with flow injection analysis was developed. It was applied to the anti-oxidation determination of maize pollen polysaccharose and natural foods. The method could be used to sieve anti-oxidation medicines.Third, a tricarbocyanine dye (Cy. 7. Cl) was synthesized and used as a new kind of near-infrared substrate for horseradish peroxidase (HRP) in flow injection spectrofluorimetric determination of hydrogen peroxide (H2O2). The near-infrared Cy. 7. Cl could effectively avoid the biological background interference.
