| The hop plant (Humulus lupulus L., Cannabinaceae) is a dioecious twining perennial. The inflorescences (hops or hop cones) are used in the breweries to give beer its characteristics such as flavor and aroma and also as herbs. On abroad, it has been used to aid digestion and as a mild sedative to treat insomnia. Chinese pharmacopoeia recorded that alcoholic extracts of hops have been used clinically in treating numerous forms of leprosy, pulmonary tuberculosis and acute bacterial dysentery.Photo-estrogen is a kind of natural chemistry material that extracts from the plant,which is used as natural substitute of hormone replacement therapy(HRT). Photo-estrogen acts for the therapy, can rise the function of the estrogen, have the extensive latent function to the related symptom of estrogen , can also prevent from the occurrence that the estrogen responds badly at the same time.This paper aimed on the comprehensive utilization of hops, the comprehensive utilization of hop, mainly engaged in the extraction technology isolation and determination of the botanic oestrogenic substance from hops. The main results were as following:(1)The contrast analyzed the fragrant type and a bitter flowers in the lupulone content, the result shows: lupulone content in bitter flowers is obvious higher than in the fragrant type.(2)Determine lupulone by RP-HPLC,with methanol:water(85:15 V/V, H3PO4 adjusts the pH2.4) as mobile phase,UV examination wave-length is 280nm. Y=10337X-5939.6 , r=0.9999.The average recover is 98%-102%.(3)From the solvent,the condition etc. and other aspect to study on totechnology of extraction.Use the single factor experiment and orthogonal test to research on the extraction condition, result shows that methyl alcohol density, temperature, time and the ratio of material to liquid all effect on the the yields of lupulone.Among them the methyl alcohol density and the temperature are most notable. (4) Using ultrasonic to extract lupulone. Use the single factor experiment andorthogonal test to research on the extraction condition, result shows that methyl alcohol density,requency of ultrasonic, times and the ratio of material to liquid all effect on the the yields of lupulone.Among them the methyl alcohol density and the times are most notable. The optimum extraction condition: the ratio of methanol is 95%, and extract for three times.The yields of photo-estrogen is 92.72%.(5)The melting agent extracts the method, lupulone purity is 56.85%, recovery rate is 68.26%.(6)Isolation by silica column:a - acid, lupulone purity is 78.95%, recovery rate is 76.56%.(7)Isolation by acrylamid column: lupulone purity is 68.85%, recovery rate is 53.86%.(8)Isolationg by acid and alkali: lupulone purity is 91.32%, recovery rate is 85.13%. |
