This paper study on the extract technology of polysaccharides in Horttuyia cordala Thunb and Litthocarpus poldstachrch Rehd. The single factor of extract time, the ratio of solvent to material, and extract times have been studied and the mathematical models have been established by using the 23 factorial experiment, and the best extracting conditions were found. The best extracting conditions of Horttuyia cordala Thunb polysaccharides is : extract time X1=2.65h , extract times X2=19.7, the ratio of solvent to material X3=26.28:1 and the yield was up to 2.61%;The best extracting conditions of Litthocarpus poldstachrch Rehd polysaccharides is : extract time X1=3.20h, extract times X2=3.01,the ratio of solvent to material X3=19.27:l and the yield was up to 3.4%.Through lots of chemic methods and instrument analysis, we study characteristics of Horttuyia cordala Thunb polysaccharides and Litthocarpus poldstachrch Rehd polysaccharides. The results indicate: Horttuyia cordala Thunb polysaccharides is a neutrial polysaccharide composed by furanside monosaccharide,and monlecular weight is 38000Da;Horttuyia cordala Thunb polysaccharides is a neutrial polysaccharide composed of four monosacchaide that is xylise,arabionse, glucose and galactose,and the proportion of four monosaccharide is 2.21:1:2.79:2.65 . Litthocarpus poldstachrch Rehd polysaccharides is an neutrial polysaccharide composed by furanside monosaccharide,and monlecular weight is 23000Da;Litthocarpus poldstachrch Rehd polysaccharides is composed of four monosacchaide that is Fructose,arabionse, glucose and galactose,and the proportion of four monosaccharide is 1:2.28:1.05:2.53.The results of Oxidation with periodate and Smith degradation indicate: Horttuyia cordala Thunb polysaccharides and Litthocarpus poldstachrch Rehd polysaccharides both have three types of polysaccharide bond that is 1â†'6 (or non deoxidized terminal), 1â†'4 (2) and lâ†'3.The antioxidant efectiveness of Horttuyia cordala Thunb polysaccharides and Litthocarpus poldstachrch Rehd polysaccharides were studied and compared with Vc usingthe superoxide radical system, hydroxyl radical system and system of DPPH. The experimental results show: Horttuyia cordala Thunb polysacc-harides has diferent scavenging efect in these radical systems. Its scavenging capability to superoxide radical is not abvious.Within the low concentration, the reduce power of Horttuyia cordala Thunb polysaccharides is less than 50% but to hydroxyl radical and DPPH is significant and its approximate IC50 is 1.35mg/mLand 1.37mg/mL. Within the low concentration, the reduce power of Horttuyia cordala Thunb polysaccharides to superoxide radical is not abvious is less than 50% but to hydroxyl radical is significant and its IC50 is 1.54mg/mL Within the low concentration, the reduce power of Horttuyia cordala Thunb polysaccharides to DPPH is less than 40%.
