The main job of this paper is to optimize the extraction process of poriacocos polysaccharide(PPS) and to purify and isolate the PPS . The results of research are shown as the following:1) Optimum process of extracting polysaccharide of poria cocos(Schw.) wolf by hot water:The optimization process conditions of extracting temperature, ratio of solid-liquid,the duration of time and the extracting times in extracting polysaccharides from poria cocos(Schw.) wolf ,has been undertaken by orthogonal experiment of L9(34).And optimum technological conditions areas follows:temperature 80°C,the solid-to-liquid ratiol:40 ,the extracting time 3h and the extracting times 2.2 ) Optimum process of extracting polysaccharide of poria cocos(Schw.) wolf by enzymolysis with hot water extraction.The result of experiment shows that the PPS extracting technology that combines enzymolysis with hot water extraction can improve the extracting rate of pachyman significantly, which is 2.32 times of that of hot water extraction. Additionally, the combined extracting technology has the advantages of shortening extracting period, milder extracting conditions, stable stereoscopic structure and high biological activity of extracted PPS.The paper also determined the optimum processing conditions for PPS extraction with the combined extracting technology: enzymolysis temperature 48°C,the solid-to-liquid ratio 1:10, enzymolysis time 90min ,the enzymolysis liquid pH value:5.0 and the amount of enzyme added i%. And after enzymolysis, hot water extracting tempreture 80°C, the solid-to-liquid ratio 1:30,the extracting time 90min and the extracting times 1.3)The examination on the concentration ratio of leaching liquor and needed alcohol quantity:In the process of ethanol precipitating, throgh examinatting the quantity of sedimented polysaccharides in the different concentration ratio of leaching liquor and needed alcohol quantity, finding that the best concentration ratio of leaching liquor (hot water extracting)and needed alcohol quantity is 1/10 time and 80%, the sedimentation is 82.03%.And the best concentration ratio of leaching liquor (combined extracting)and needed alcohol quantity is 1/5 time and 80%, the sedimentation is 83.15%.4) The comparison of the methods of eliminating proteinThrough comparing the trichloroacetic acid method,Sevag method and the combination of enzymic method and Sevag method by the elimination ratio of protein and the loss ratio of polysaccharide.Finding the best way to remove protein from PPS is that: the combination of enzymic method and Sevag method.5) Research of the purificationg PPS by using macroporousion-adsorption resinDuring the purification process with column chromatograghy,nine types of macroporousion-Adsorption resin were compared and determine the best one was AB-8 .And the operating conditions on elution were established. The best eluting solution is 0.2mg/ml NaCl .The best velocity of elution is 2ml/min.6) Purify and isolate PPS by DEAE-cellulosePurify PPS detach by DEAE-cellulose pillar.Using the different solution to wash the pillar. Using the anthrone-sulphoacid method to test the content of polysaccharides. We can get one neutral polysaccharide PPS1 and three acid polysaccharides PPS2,PPS3,PPS4.7) Test the monosaccharides composition of PPS2Through the analysis of GC of PPS2,finding that PPS2 is composed with the rhamnose,seminose,galactose,glucose,gum sugar.8) The research of development and utilization of PPSMake the PPS tablets.Determine its prescription and technological process.
