In this thesis, through extracting fruit of piper longum L. with ethanol and petroleum ether, three extracts such as ethanol extract I , Petroleum ether extract, ethanol extract II were obtained. Piperine was preparated with ethanol extract II by recrystaling. Petroleum ether extract and ethanol extract I were farther analyzed. Six fractional-F6) were isolated from petroleum ether extract by utilizing column chromatograph, TLC techniques. F3 was sifted from six fractions through experiment of antilipidic effect for male Wister rats. F3 can effectively reduced serum TC in rats.Three ingredients(F3-0, F3-1, F3-2) and a component(GBR-7) were isolated from crude F3 by utilizing column chromatograph, TLC, recrystaling techniques. A high-pressure liquid chromatograph with the Waters 600E pump, the Waters2487 UV detector, 300x3.9mm i-d. Symmetry C18 column, 300xl9mm i.d. SymmetryPrep Ci8 column was used. Mathenol-water was used as the eluting system. Through monitoring the column effluent at 254nm, twelve components (3-A-3-L) was isolated from crude F3. Through detected by utilizing HPLC, the ingredient(F3-l) isolated from F3 through methodes of column chromatograph and TLC was consist of 3-A, 3-B, 3-D, 3-H. F3-2 isolated from F3 through the samemethods was consist of 3-G, 3-J, 3-L. GBR-7 and 3-D was the same compound.The structures of five components were identified by MS, 1HNMR, 13CNMR & its DETP, HMQC, HMBC, COSY of 2DNMR data and other chemical data. The complete structures of three compound were identified from there spectral characteristics as (E,E)-N-(2-methylpropyl)-2,4-decadienamide (GBR-4),(E,E,E)-11 -(1,3-benzodioxol-5-yl)-N-(2-nmthylpropyl)-2,4,10-undercatrienamide(GBR-5), (E,E,E)-13 -(1,3-benzodioxol-5-yl)-N-(2-nmthylpropyl)-2,4,12-tridecatrienamide(G BR-7). The others were inferred as alkene with high symmetrical structure.Further antihyperlipidenic experiments show: F3-1 isolated from F3 can reduce serum TC, TG, APOB.raise serum HDL-C in rats. F3-2 isolated from F3 can effectly reduced serum TC, TG, APOB in rats, and elevated serum HDL-C in rats. Compared with Simvastatin, F3-2 was higher antiperlipidemic effect.
