The extraction and separation of rutin and glycyrrhizic acid which were the active constitution of Sophora japonica L. and Glycyrrhiza uralensis Fisch. were studied in this dissertation respectively.The flavonoids was extracted from Sophora japonica L. by heating-reflux and ultrasonic extraction. The optimum conditions of ultrasonic extraction was obtained by orthogonal design. Effects of experimental conditions on the extraction rate and content of flavonoids were discussed in detail. Under the optimum conditions, the average extraction rate and content of flavonoids were 22.28% and 92.09% respectively. Compared with heating-reflux extraction, the extraction rate and content of flavonoids prepared by ultrasonic extraction were obviously increased. This showed that Ultrasonic extraction was much better than heating-reflux extraction. The concentration of flavonoids and the purity of rutin and quercetin were determined by spectrometric method and HPLC. The polyamide adsorptive resina has perfect adsorption and desorption effect on the flavonoids of Sophora japonica. The purity of rutin and quercetin which were purified by column chromatography were 96.64% and 96.28% respectively. The chemical structure of rutin and quercetin were determined by FT-IR,MS,~1H-NMR. The single crystal of rutin was determined by X-ray single crystal diffraction. The coordination complexes were synthesized at the experimental conditions optimized. Rutin and quercetin were selected as organic ligand, the complexs of rutin-Cu(Ⅱ),rutin-Zn(Ⅱ),quercetin-Cu(Ⅱ),quercetin-Mn(Ⅱ)were synthesized and the chemical structures were characterized by FT-IR.The glycyrrhizic acid was extracted from Glycyrrhiza uralensis Fisch. by ultrasonic extraction. The optimum conditions on the method were obtained by orthogonal design. Effects of experimental conditions such as solvent,concentration of solvent,ultrasonicpower,ultrasonic time,meshe on the extraction yield of glycyrrhizic acid were discussed in detail. The concentration of glycyrrhizic acid in the extract of liquorice was determined by spectrophotometer.
