Lipase is important in the bio-chemical and it is widely using in Biodiesel production recently. But the price of the lipase using in Biodiesel production is too high. So we can screen for lipase-producing bacterium to reduce the price of Biodiesel.Collecting 20 shares soil around the brew house, flour factory, gas station, grain shop and cultured in abio-medium added olive oil. Screen in olive-PVA emulsion flat, and find NO.22 bacterium that have high enzyme activity. By observe the form of the colonial morphology and amplification its 16s rRNA sequence, we can conclude NO.22 bacterium is Aranicola sp. .Use NO.6 bacterium to study lipase-related gene. This part is the base of construct gene-engineering bacterium. Chromosomal DNA from NO.6 bacterium was partially digested to 1-3kb with the restriction enzyme. The plasmid DNA was cut and dephosphorylated. The linearized plasmid DNA was ligated to the Aranicola sp. DNA fragments and resulting recombinant plasmids were used to transform E.coli DH5a. Now the screening of genomic library is under way.Observe the affect polar solvent to the lipase active. Added different dosage of methanol, aether, isoamyl alcohol into the medium, observe the lipase active after 72 hours. Find that methanol has depressant effect to the lipase activity, aether can enhance the lipase activity and isoamyl alcohol has no effort to the enzyme activity.
