Partâ… Determination of Rafoxanide content in food of animal originRafoxanide is a salicylanilide class anthelmintic drug,which make worms paralytic,dead to kill them through preventing fasciola hepatica from oxidized phosphorylation. Rafoxanide has been widely used to control fasciola hepatica in sheep and cattle and has good therapeutic effect.In resent years,The reports of Rafoxanide were main on pharmacology, pharmacodynamic action.There are little literatures about the determination of Rafoxanide content in cattle tissues.This thesis develops the method for determination of Rafoxanide content in tissues and fits for studies on the pharmacokinetics.A HPLC method for determination of Rafoxanide content in tissues of cattle has been developed.Rafoxanide was extracted from tissues of cattle with acetonitrile.The extraction was cleaned up by passing through aluminum oxide neutral column and separation through Kromasil C₁₈(4.6×250mm,5μm)column with UV detection at 282nm.The mobile phase was acetonitrile—water(pH2.5)(V:V=83:17)at 1.5mL.min^-1and the column temperature was 30℃.There was a good linear relationship for Rafoxanide within the range of 0.05～0.4μg.mL^-1(r=0.9999),the average recovery at three concentration levels such as 0.01,0.1, 0.4μg.g^-1were 78.3%,85.8%and 95.1%in muscle;73.3%,83.9%and 91.9%in liver;74.3%, 81.9%and 92.8%in kidney respectively.Partâ…¡Synthesis and process research of 2,6-dimethyl-β-cyclodextrinβ-cyclodextrins are cyclic oligosaccharides consisting of seven(1â†'4)-linked D-glucopyranosyl residues,the size of their molecule holes are suitble and cost of production is low,but the solubility in water ofβ-cyclodextrins is low resulted from hydroxyl group linked by intramolecular hydrogen bond between C-2 and C-3 ofβ-cyclodextrins,which limits their application in the field of factory.The solubility in water of methylatedβ-cyclodextrins is larger,toxicity is low,the cost is lower and has been widely used.In resent years,Research about chemically modifiedβ-cyclodextrins was main on methylatedβ-cyclodextrins.Therefore,we research its synthesis and process. Usingβ-cyclodextrins as the starting materials,dimethyl sulphate as methylated reagent, dimethylformamide and dimethyl sulphoxide as reaction reagent,in basic condition by barium hydroxide octahydrate 2,6-dimethyl-β-cyclodextrin is prepared.
