The modification of silica-based material was a vital step in the efficient separation and analysis of biological samples by HPCE and HPLC.And therefore occupied a decisive position in the research of High Performance Capillary Electrophoresis(HPCE) and High Performance Liquid Chromatography(HPLC).The silica-based capillary and micro-sphere were modified by the multi-hydroxyl compoundsγ-Glycidoxypropyltrimethoxysilane andδ-Gluconolactone,their application in HPEC and HPLC were investigated in this thesis.The obtained results were as follows:1.The capillary was prepared by covalently bondingγ-GPS onto the inner surface according to the literature method.The electroosmotic flow(EOF),basic proteins' separation efficiency and stability of the column were all investigated.The results showed the mean separation efficiency for three basic proteins are 150000 plates/m,the mean standard relative deviation(R.S.D)referring to the protein retention time of intra-day and inter-day is 2.7%and 4.6%,respectively,and the reproducibility was in the range of 90.4%-94.7%.2.δ-Gluconolactone was covalently coupled to aminopropyl derivatized capillary,which created hydrophilic brushes on the inner wall of the capillary.The coated capillary was shown to generate a stable electroosmotic flow(EOF) in the investigated pH range of 2.0-9.0 and to suppress effectively the adsorption of proteins.And it enabled separation of some biopolymer mixtures including basic proteins,DNA and tryptic digested bovine serum albumin(BSA) within 15 min with efficiencies up to 450,000 plates/m.The intra- and inter-day reproducibility of the coating referring to the retention times of proteins were satisfactory with mean relative standard deviations(RSD) of 0.8 and 1.4%,respectively.3.New hydrophilic chromatographic stationary phase was prepared by bondingδ-Gluconolactone onto the surface of silica.Six herb components with strong polarity were separated by hydrophilic interaction chromatography mode,and the effect of mobile phase,salt concentration as well as pH value on the retention behavior of solutes were investigated. The results indicated that six strong hydrophilic herb components could be separated by the new column under the mode of hydrophilic interaction chromatography,and in the separation of the Danshensu injection was also carried out with the satisfactory efficiency.
