An important medicine intermediate Ethyl 5-Nitro-4-oxopentanoate was synthesized firstly using succinic anhydride and ethanol by the preparing of monoethyl succinate,ethyl succinyl chloride and Ethyl 4-(Imidazol-1-yl)-4-oxobutyrate.The products were characterized by spectrum of IR,1H-NMR,GC,MS.Effect of the reaction temperature,reaction time,reactants ratio,catalyst and solvent on the reaction was investigated through single variable experiments.The optimum reaction conditions were obtained.The yield of monoethyl succinate was 83%when the mole ratio of ethanol and succinic anhydride was 2:1,reaction temperature was 80℃,reaction time was 4h and the ratio of resin to succinic anhydride was 30%.Being stored for only one month,the white deposition in the pure monoethyl succinate appeared,and the white deposition was identified as succinate by methods of physics and chemistry.Then the reason was found out and the storage was improved.By using the solvent of THF,the yield of Ethyl 4-(Imidazol-1-yl)-4-oxobutyrate was 92%when the mole ratio of imidazol and ethyl succinyl chloride was 2:1,reaction temperature was 30℃,reaction time was 4h.By using the catalyst of strong alkali t-BuOK,the yield of 5-Nitro-4-oxopen-anoate was 80%when the mole ratio of nitromethane and 4-(Imidaz-ol-1-yl)-4-oxobutyrate was 1.5:1,reaction temperature was 30℃, reaction time was 48h and the mole ratio of t-BuOK and 4-(Imidazol-1-yl)-4-oxobutyrate was 2:1.
