β-lactam antibiotic is a kind of antibiotics withβ-lactam ring in chemical structure,as to theβ-lactam antibiotics with powerful antibacterial activity,broad antimicrobial spectrum and low toxicity,it has common application in animal breeding process for preventing and treating animal diseases.While with its The long-term abuse and irrational use,will cause human allergy and bacterial resistance,as well as drug-resistant strains of adverse impact on the environment.thereby,will be a serious threat to human health.So it is urgent to establish the detection methods ofβ-lactam antibiotics drug residues,so as to protect the safety of food and the health of humanbeings.There are four parts in the thesis.The first chapter describes the structural characteristics,physical and chemical properties,pharmacological action and residual hazards in animal food of theβ-lactam antibiotics;reviewe the recent developments ofβ-lactam antibiotic residue analysis by Liquid Chromatography,Gas Chromatography,Thin-layer Chromatography,Chromatography-Mass Spectrometry,radioactive receptor analysis and other modern analytical techniques and sample treatment techniques in animal foods.The second and The third chapters establish assay method of 13 kinds ofβ-lactam antibiotic residues such as penicillin G,penicillin V,amoxicillin, carbenicillin,ampicillin,oxacillin,cloxacillin,nafcillin,penicillin G,penicillin V, amoxicillin,carbenicillin,ampicillin,oxacillin,cloxacillin,nafcillin,cefquinome, cephalexin,cephradine,cefazolin and cefoperazone in animal food such as pigs meat, pork kidney and milk by Ultra High Performance Liquid Chromatography-tandem quadrupole mass spectrometry(UPLC-MS/MS),use of UPLC-MS/MS high sensitivity and separation speed,while the advantages of qualitative and quantitative. Baker C18 was used to extract,purify and isolate thirteenβ-lactam antibiotics in pork,pig kidney and milk.,0.1%Formic acid-Acetoni and 0.1%Formic acid-water solution as mobile phase for gradient elution,ionization was producted by ESI+ and determined with MRM model.For all of the thirteenβ-lactam antibiotics,the limit of detection were 1μg/kg,the limit of quantification were 2μg/kg,which could realize high speed qualitative and quantified analysis of trace residues for all the thirteenβ-lactam antibiotics in 4 minites. The forth chapter establish assay method of melamine residues in eggs by UPLC-MS/MS in the first time,Retention time was 0.8 minites,the limit of detection was 5μg/kg,the limit of quantification was 10μg/kg.Method is rapid and simple, specific and sensitive for the detection of melamine in eggs.
