Extracellular polymeric substances (EPS) are heterogeneous biopolymers produced by Gram-negative and Gram-positive bacterial cells. They are hydrated polymeric molecules around biofilms. The main components of EPS are polysaccharide,protein,nucleic acid,fatty acid,uronic acid and so on. The composition and physico-chemistry properties of different bacteria are different which contribute to the biosorption,biofilm structure and biology characteristics.There are a few studies on EPS of bioleaching bacteria in these years. During the bioleaching process, the effect mechanism of EPS still needs to be studied.This paper studied on the growth characteristic and oxidation ability of HQ0211 bacteria.EPS of HQ0211 bacteria in the Logarithmic phase were extracted by four methods. The concentration of polysaccharide and protein were 0.1873mg/mL and 0.1298mg/mL when the concentration of bacteria was 104-105 cell/mL.So we can conclude the content of polysaccharide is more than the protein. Through research and choice of four different methods, glutaraldehyde, EDTA, EDTA(buffer), H2SO4, EDTA method can be drawn in terms of other means exist for better stability. The research reveals what influences the results of extraction are the growth of bacteria, activity, external growth environment, extraction reagent as well as extraction methods, etc.This paper researched the influence of carbon on bacteria growth. From the experimental results we can see that the appropriate concentration of charcoal and coke played a role for bacterial growth and accelerated the growth of bacteria.It entered the logarithmic growth phase earlier than that the bacterial merely in 9 K culture medium.The adsorption to minerals of HQ0211 bacteria was studied in this experiment. By measuring cracking time, wavelength, cooling time,the optimal conditions are the best cracking time is 25 minutes,the best wavelength is 430 nm, the best cooling time is 10 minutes.The adsorption capacity,dissociative capacity of HQ0211 bacteria under different Eh was determinated under optimal conditions.Experiment reveals that the adsorption bacteria account for the total bacteria relationship with the potential changes.Finally,this paper also discusses the dynamics of bacterial adsorption process.
