With the development of computer science and geographical information science, geographical information system has been used widely because of its strong spatial analysis function.Route analysis is the most general and basic function in GIS. Its key content is the optimum route analysis. In this paper, the main content was the application of optimum route analysis to the public transportation. The whole framework is based on establishing model, studying algorithm and achievement. According to the network model of public transportation, the author analysed the bus stations' distribution and abstracted it, then expressed the public transportation network with virtual network. Traditional Dijkstra algorithm and some relevant improved algorithms are based on the shortest route, but in fact the least transfer is the most important when passengers go out. Based on the least transfer, an algorithm was brought forward. The dynamic segmentation is a new technology about management linear factor, which has been realized in ARC/INFO. Some methods on how to realize the dynamic segmentation in MapInfo were discussed. Finally, an overall scheme of "Information inquiry system of urban public transportation" was brought forward, and some modules according to the real data of ChangSha have been obtained.
