Hedysarum pol-botrys Hand.-Mazz. is one of the main traditional Chinese medicine planted in Gansu province, and very widespread in Dangchang, Wudu, Min County, Zhouqu, Lintan, Zhang County, West and, in Li County, Wushan, etc. It both has wild species and cultivar and kingpin in Gansu, Guangdong, Fujian, and exports. As has been the mix of Hedysarum pol-botrys Hand.-Mazz. and Astragalus membranceus-(Fisch.) Bge., it has yet to see the research reports of Hedysarum pol-botrys Hand.-Mazz. seed quality standards, and karyotype analysis. The objective of this study were to study the seed quality standards and karyotype of Hedysarum pol-botrys Hand.-Mazz. in order to provide the necessary Cytological inform-ation to study the evolution, identification and fine seeds selection and provide a theoretical basis for further research and development and wider application on Gansu Hedysarum pol-botrys Hand.-Mazz. The main findings of this paper is as follows:1. From the germination test results, the seed germination quality improved with temperature elevation and the optimum is 25℃resulting in 67.33% of germination rate and 35.33%of germination power. However,when over 25℃, the seed germination decreased significantly. The higher temperature also restrained the sprout growth. The Hedysarum pol-botrys Hand.-Mazz.seeds rubbed with sand, soaked in H2SO4 and soaked in water after rubbed germinated at a remarkably higher rate than the seeds treated in other ways and CK and thus it followed that the mechanical inhibition resulting from the testa of Hedysarum pol-botrys Hand.-Mazz. seeds was the direct reason why Hedysarum pol-botrys Hand.-Mazz. seeds germinated at a low rate. The seeds rubbed with sand reached in 85.33% of germination rate and 35.33%of germination power,32% higher than the control of germination rate and 48% higher than the control of germination. The seed germination quality difference of different maturation seeds are very great, the germination of well maturation seed is quick and order. The seed germination rate and the dry weight of seedling of ill maturation seed fall clearly.2. Germination percentage was the primary indicator of seed quality grading, thousand-grain weight, cleanliness andmoisture contentwere important reference indicators. The seed quality of each grade should reach the following requirements:for first grade seeds, germination percentage≥85%, purity≥98%, thousand-grain weight≥4.0g, seed moisture≤9%; for second grade seeds, germination percentage 80%~85%, purity 95%~98%, thousand-grain weight 3.0~4.0g, seed moisture 9%~10%; for third grade seeds, germination percentage 70%~80%, purity≤95%, thousand-grain weight 2.0~3.0g, seed moisture≤11%.3. Staining and Slide-preparing Technique of Hedysarum pol-botrys Hand.-Mazz. is that the best experimental protocol is better when sampling time at about 10:00AM, root length to be about 1~1.5cm, the pre-treatment time with saturated p-dichlorobenzene lasts 3h, the dissociation with 1mol/L HCl is at 60℃for 13min, and the dyeing time lasts 13min with modified carbolic acid-magenta.4. The fukanensis is diploid. The chromosome number of somatic cells was 2n=14. The karyotype formula is 2n=2x=14=10m+4sm. The 4th and 7th are submetacentric with its AR is between1.71~3.00, and the others are metacentric with its AR is between 1.01~1.70. Its index of Asymmetric was 60.45%. The karyotype of Hedysarun polybotrys Hands.-Mazz. belongs to"2A"type of stebbins, and it is a primitive species. The conclusion of the study provides an important scientific basis of identification between the Hedysarun polybotrys Hands.-Mazz. and Astragalus membranaceus. The test was not observed satellite.
