Astragalus membranaceus var.mongholicus is a perennial herb of Leguminosae family.Its roots have indications for strengthening the spleen and tonifying the lung,as well as nourishing blood and engendering liquid and the effect of soring muscles.By collecting seeds samples,seedlings and medicinal materials from the main production area of A.membranaceus var.mongholicus,this study explored and formulated the grading criteria of Astragalus seeds,in order to provide an important basis for the modern mechanized development and standardized production of Astragalus.The results of this paper are showed as follows:1.Progressive screening studies have shown that the test parameters of A.membranaceus var.mongholicus seed test include sample,clarity,germination rate,weight,water content,viability and purity.The seed sample method is a quarter method,and the grain weight is preferably a thousand-grain weight method.The germination conditions were preferably germinated on a 15 ° C filter paper bed after soaking for 18 h,the germination was started on the second day,and the germination was terminated on the 7th day.The water content of seeds was determined by drying method.It was the best to dry at 105 °C for 6 h.The seed viability was determined by TTC method.It was best to soak the cotyledons in 0.5% TTC for 30 hours at 30 °C in the dark.The purity uses protein extraction by the mercaptoethanol method,and it is preferably separated by polyacrylamide electrophoresis(SDS-PAGE).2.According to the difference screening test on seed characteristics,the standard indicators of seed quality classification of A.membranaceus var.mongholicus have germination rate,1000-grain weight,viability,water content and clarity.The 60 seeds of A.membranaceus var.mongholicus collected in the national production area were divided into 3 grades,that is,the germination rate of first-class seeds is ?75%,the level ?55%,the third level ?35%;the first-class seed 1000-grain weight ?6.90 g,the second level is ?6.30 g,the third level is ?5.70 g;the first-class seed viability is ?80%,the second level is ?70%,and the third level is ?60%.The grades of the three grades were all ?95%,and the water content was ?10%.3.The differential screening test on the seedling characteristics index showed that the standard indexes of the quality grade of the seedlings of A.membranaceus var.mongholicus were root length,root thickness,weight per plant and root vigor.The 36 seedlings of A.membranaceus var.mongholicus collected from the national production areas into 3 grades,namely: first-class seedling root length ?35.0 cm,root thickness ?4.7 cm,single plant weight ?3.4 g;second-class seedling root Length ?29.0 cm,root thickness ?4.2 cm,weight per plant ?2.4 g;root length of third seedling ?23.0 cm,root thickness ?3.8 cm,weight per plant ?1.4 g;root activity of different grades of seedlings OD485 nm ? 0.900.4.The difference screening study of the characteristic indexes of medicinal materials showed that the standard indexes of quality grades of A.membranaceus var.mongholicus were main root length,root length,root diameter,single weight,lateral roots number,disease grade,moisture,ash,leaching,Astragaloside IV and Calycosin-7-O-?-D-glucopyranoside.The 41 A.membranaceus var.mongholicus were divided into 4 grades,that is,each medicinal material grade index was: the first ‘s root length ?60 cm,root diameter ?15 mm,single root weight ?65 g;the second's root length is ?45 cm,the root diameter is ?11 mm,the single root weight is ?38 g;The the third's root length is ?30 cm,the root diameter is ?7 mm,the single root weight is ?11 g.Four-class: general goods.Regardless of root length,root diameter and single weight,the content requirements in accordance with the Pharmacopoeia regulations can be.5.The fingerprints of quality characteristics of A.membranaceus var.mongholicus were established by HPLC-UV method.The standard products were used to identify eight major components of Calycosin-7-O-?-D-glucopyranoside,Calycosin,Ononin,Formononetin,Astragaloside IV,Astragaloside?,Astragaloside II,Astragaloside ?.System clustering Analysis of 21 Astragalus from 7 producing areas was divided into 3 categories,the average composition of various medicinal herbs was 94.43%,indicating that the chemical composition of 21 parts of A.membranaceus var.mongholicus in different producing areas is consistent,which is consistent with the characteristics of A.membranaceus var.mongholicus.
