Lycium barbarum L. (Solanaceae,Lycium), ranks the first of the five sorts of treasures or the local famous special products in Ningxia. It has peculiar effects on nourishing liver, kidney, making one's eyes bright and enhanced male genitals. Up-to-date studies have shown its effects on anti-cancer and resisting caducity. Lycium barbarum L. is a perennial deciduous bush in the solanaceous family, and is an important tree for commerical fruit growing in Ningxia areas. With the further development for medical use and health care recently, the cultivated areas have increased continuously. The convential breeding cannot meet the requirements for Lycium barbarum L varieties improvement. At present, haploid plants through anther culture of haploid breeding generated the double haploid plants, can shorten the breeding period, simplified selection process. The study of Lycium barbarum L.'s anther culture only remain in the theory, and not for the production, led to the slow pace of Lycium barbarum L. breeding. This study research the effects of Lycium barbarum L. anther culture (for example: basic medium, hormones, activated carbon, temperature, mannitol pretreatment) systemly. The purpose is getting regenerated plants, establish a more suitable system of Lycium anther culture, to enhance breeding efficiency of Lycium barbarum L., quicken the speed of Lycium barbarum L. breeding selection,Four genotypes of Lycium barbarum L. were used to study the effects of factors on brown anthers, callus induction and embryoid yield in the orthogonal array of L32(2×45), including Ningqi No.1, Damaye, No.0207, Mengqi No.1. The results were as follows:1. There were significant roles in anther response to culture in different genotypes. Obvious difference was observed in 4 genotypes inoculated for brown rate, callus induction, and embroyoid yield. Ningqi No.1 was easy to be induced. On the contrary, 0207 was difficulty to be induced.2. To two culture medium, MS is suitable for anther culture of Ningqi No.1 and Damaye, and induction rates of the anther of 0207and Mengqi No.1 on B5 were higher than MS.3. Phytohorm one played an important role in Lycium barbarum L. anther culture. The medium added with 1.0mg/L 2,4-D and 1.0mg/L KT was suitable for the induction of callus;4. In three kinds of tempreture-treat, bud 4℃pre-treat and 32℃training one day, then transferred to 25℃drak culture, this kind is suitable for Ningqi No.1.5. Medium added with 0.8g/L active carbon could reduce the anther brown rate, and improve the rate of embryoids effectively, but decrease the rate of callus induction.6. Anther were pretreated with mannitol, and the callus induction rate was increased, the suitable concentration was 0.3mol/L.7. The response of anther to culture was genotype dependent. Active carbon had significant impact on anther brown rate, ranking as active carbon﹥mannitol﹥culture medium﹥heat stock treatment﹥phytohormone; Culture medium and mannitol played significant roles in callus induction, ranking in phytohormone﹥culture medium﹥mannitol﹥active carbon﹥heat stock treatment; Active carbon, culture medium and heat stock treatment influenced seriously on embryoids, ranking as phytohormone﹥active carbon﹥culture medium﹥heat stock treatment﹥mannitol.
