In this study, Lycium barbarum var qingqi1 was treated as the research object.Different explant disposal, blade callus induction, callus multiplication,callus differentiation, stem adventitious bud induction and rooting culture were setted as 6 periods.The growth status in every period was studied to analyse and summarize the best treatment efficiency, and explore the best hormone combination.The result showed that:the best selecting materials time isMay and June.May and Juneare the best time for sampling. At this period,the young sprouts growed exuberantly, and the toxic substance accumulated less; The best concentration of HgCl2 is 0.10mg/L,the best suitable time of disinfection is 10 min.It could kill the bacteria and would not hurt the explants.The best hormone combination of leaf blade callus induction is 6-BA0.7mg/L+NAA0.7mg/L.They had highly altitudinal redifferentiation ability and exuberant multiplication ability. The best hormone combination of callus multiplication is 6-BA1.0mg/L+IBA0.8 mg/L +NAA 0.2mg/L.It reached the propagation fastigium in the 15 th. Callus volume increased about 5cm3. They have higher differentiation ability. The best hormone combination of callus differentiation is KT1.3mg/L+NAA0.02mg/L, the leaf primordium began to appear in the 6th, a lot of leaf primordiums appeared in the 9th,the differentiation rate was 86.7% and the buds were exuberant. The best hormone combination of adventitious buds induction is 6-BA0.8mg/L+NAA0.05mg/L,the induction rate was 86.7%, the length of average bud was 5.83 cm, and the new buds was exuberant.The best hormone combination of rooting culture is IBA0.3mg/L+NAA0.3mg/L,it was one week that the root began to appear,the rooting rate was 88.3%, the root and bud were exuberant.
