Wall for the first time the United States stem from extracts of camptotheca isolated camptothecin(CPT), then people found that CPT on gastrointestinal cancer, bladder cancer, liver cancer and leukemia and other malignant tumors have a certain effect. The unique mechanism of anticancer activity of CPT launched a study of CPT in camptotheca acuminata and the climaxThis paper carried out research on isolation, screening of endophytes of CPT from Zhangjiajie and studies antitumor acitivity of endopyte product. We aim to exploit new sources for production of CPT, isolate endophyte strains for CPT exploit bio-active natural products and provide technology support for industrial production of CPT.CPT unique mechanism of antitumor activity of camptothecin and CPT up the climax of study. A new environment-friend method to produce CPT is realized, using endophytic fungi isolated from the cortex of camptotheca acuminate decne.15 strains of endophytic fungi were isolated,then cultured in the liquid potato medium and indentified by means of the thin layer chromatography (TLC) and high performance liquid chromatographic (HPLC). The result showed one strain of fungus which was signed EFC 1-03 could produce CPT and this strain has further research value.Sending the EFC 1-03 strains into the outer-space, researching the influence of space breeding to the physiological characteristics and the ability of producing camptothecin of endophytic fungi isolated from camptotheca acuminate. Results showed space mutagenesis could make the first generation strains growing faster, but this advantage can't be geneticed, and it didn't improve endophytic fungi's ability of producing CPT.Adding KH2PO4 MgSO4 and (NH4)2SO4 into the medium, using three factors three levels orthogonal experiment optimizing the medium, results shows using NO.4 scheme can obtain the best effect, getting the highest production of CPT.
