Cordyceps. is an important medicinal fungi and widely used in China for thousands of years. In taxonomy, it belongs to Ascomycetes Hypocreales Ergot Division Cordyceps. The fungus is endemic in the alpine zone of high mountains with cool weather conditions and have a low yield, the medicinal value is very broad, especially in anticancer. Now, Cordyceps become more famous with more researchs about its efficacy, but its lab culture and molecular modification has not be reported. So, the research to increase its medicinal value will very important and have ecnomic prospect.The structure and function of fungal immunomodulatory protein—a new protein family,are like to immunoglobulin and lectin. The first FIP called LZ-8 was purified from Ganoderma lucidum by kino et al at 1989, and other family members were found one by one. It can promote proliferation of human peripheral lymphocytes cell and rat spleen lymphocytes cell, enhance function of immunity.According to this research, Cordyceps was isolated from the body of larvae collected from xinjiang for morphological and DNA moleculor identification, and the reasult prove that the mycelium is Cordyceps scarabaeicola. To receive a purified and thrived fungus, the culture condition were improved.The Fip gene was amplificated from Ganoderma lucidum by RT-PCR, and the ER retention signal was used to improve the expression level of foreign protein. On the basis of previous reported, we chose GPD promoter isolated from L.edodes, with bialaphos resistance as the selective marker, to construct expression vector—p3300bfipk. through Agrobacterium-mediated transformation method, we transformed the vector into Cordyceps scarabaeicola, and obtained a lot of resistant fungus. PCR was done to find transgenic fungus, furthermore RNA was extracted by TRIZOL, and RT-PCR was done to check the expression on transcription level in some positive fungus.In this study, molecular modification in Cordyceps scarabaeicola was done, it is a new way to research Cordyceps a traditional medicinal fungi, and provide a certain experimental basis to appile to actual.
