As a kind of well-known medicinal fungus, Ganoderma lucidum has been proved that it is an effective therapeutic adjuvant for cancer, chronic bronchitis, coronary heart disease, hepatitis, high blood lipid disorders, neurological disease and so on to certain degree. Another edible and medical fungus Flammulina velutipes, can not only prevent and cure hepatitis & gastrointestinal ulcers, but also decrease serum cholesterol. It is hard to cultivate these two fungi with a traditional method wasting time and efforts, and to control the quality. A new technique for a large-scale production of these two fungi is the liquid submerged fermentation. The advantages of this new method are obvious, such as higher yield, less place, shorter time, and more-easily controlled. The micro-protein, fungal immunomodulatory protein, extracted from G. Lucidum and F. velutipes with the immune-regulated function has biological activity including agglutinating blood cells, promoting lymphocyte proliferation, and inducing immune cells to secrete cytokines.This paper preliminarily studies the liquid-submerged fermentation process of the velutipes mycelia of F. velutipes and G. Lucidum. The biological activities were tested towards different cultivation on peanut and soybean medium respectively, and the growth status in shake-flask and stationary cultivated plates was recorded. The result was that it could get higher protein and sugar content totally of F. velutipes mycelia in the peanut-medium cultivation by stationary cultivated plates. It could get the higher dry weight and fresh weight of mycelium in stationary cultivated plates than in shake-flask.It demonstrated that the recombinant fungal immunomodulatory protein might be the glycosylated protein in the yeast expression system, which was analyzed by the Phenol-sulfuric acid method. Also the purified recombinant protein has a high purity qualify through an optimized recombinant expression technique. The biological activity test also showed that the purified recombinant fungal immunomodulatory protein could agglutinate blood cells and promote lymphocyte proliferation in mice.This study optimizes both the submerged fermentation media of G.Lucidum and F. velutipes and the recombinant expression technology of funal immunomodulatory protein. Meanwhile it provides an experimental evidence for the submerged fermentation of G. Lucidum and F. velutipes, and a theoretical foundation for the further study on fungal immunomodulatory protein.
