In this article, we clone the 3?untranlation region of actin gene family in BPH by improved 3-RACE mothed and sequence the cDNA clones what we get. Basing on results we had got ,we test expression of every clone in deferent development stage of BPH by RT-PCR. Integrate the sequences message and results of RT-PCR test; we make a tentative determine of the nature of every cDNA clone. In the experiment of 3-RACE, Three down-stream primers all can amplification band. By contract, the best primer is 0422-7,and we select it as the down-stream primer in the RT-PCR test. As actin gene family is conversation in protein coding region but not in 3? untranslation region. So from what we study we can conclude that there are five actin genes which named BPH-A,BPH-B,BPH-C,BPH-D,BPH-E by us in BPH抯 actin gene family; and divided them to three sub-groups sub-group 111111 accord to their characters in sequence and expression in different stage of BPH. In the sub-group II First, we can view a gene expression regulate at the level of post-transcription: after the niRNA抯 transcription of BPH-B it can polyadenylation in three deferent polyadenylation signal position and product three different mRNA. In the RT-PCR test, they express standly in every development stage of BPH ,and at 61 the specific amino-acid position of their protein terminal the same as any other cyto-actin gene which had been view clearly, so we think they are cyto-spcciflc actin gene.To the BPH-A it express only from 3 rd nymph to adult stage and at the specific amino-acid position of their protein terminal it has the same amino-acid as any other muscle-actin gene so we conclude it is muscle-specific actin gene needy in post- nymph stage and in adult stage.To the BPH-D, it expression only in nymphet stage and at the specific amino-acid position of their protein terminal it has the same amid-acid as any other muscle-actin gene so we judge it is muscle-specific actin gene needy only in nymph stage.
