AbstractThe better way of extracting genomic DNA from tobacco was found, and effects and procedure optimum of RAPD in tobacco have also been studied. With the RAPD technique, we got the RAPD fingerprints of the 52 flue-cured tobacco cultivars. The analysis result showed that variations were widely existed in the 52 flue-cured tobacco varieties, and the dendrogram was constructed for them. We also have studied the genetic accordance in the 5 individuals of the same cultivar. The result showed that the different individuals of the same cultivar were very similar in genome, the fingerprints which were got in the experiments, could be applied on the works such as identificating cultivars and detecting the fake seeds and so on. With the RAPD technique, we also have found that the RAPD marker S95750 was close linked to the black shank resistant gene, and S220830 was close linked to brown spot resistant gene.
