| ABSTRACT Several insecticidal genes such as modified cowpea trypsin inhibitor (SCK) gene, modified Bacillus thuringiensis (B.t.) gene and both the SCK and B.t. gene had been successfully transferred into a maize elite inbred Rl8. A large number of fertile transgenic plants had been obtained. Some conditions such as transformation manipulation hormone, genotype medium , culture conditions, selection agents, selection manners, subculture time and methods of transplant had been studied and optimized in the experiments. Using all the methods mentioned above, an efficient and stable transformation system of embryogenic calli of elite inbred line R18 was established. Results of PCR, PCR Southern, Southern blot, B.t. ELISA, Typsin remain vitality and bioassay of transgemc plants confirmed that transgenic plants carry the foreign genes, and express the target genes in defferent degree. Bioassay of transgenic maize showed significantly increased resistance to the larvae of maize Hubner(Ostrinia palustralis) than untransformed control plants. By now, T1 seeds had been obtained, which would pave the way for further maize agronomic improvements through genetic engineering. |
