FtsZ (filamentation temperature-sensitive, fts) is a functional protein in prokaryote and higher plants, which controls the division of prokaryotic cell and plastid in plants. It is important for us to study the gene expression and regulation of FtsZ gene, which will provide a new pathway to learn further many important problems, such as the molecular mechanism of prokaryotic cell, plastid division and the origin and evolution of eukaryotie cell. In order to study its gene expression, regulation and localization, a FtsZ cDNA isolated from Nicotiana tabacum was constructed into an expression vector fused with glutathione S-transferase (GST). After transforming into E. coil strain JM1O9, the GST-FtsZ fusion protein was induced to overexpress by IPTG and purified with glutathione-agarose affinity chromatography and preparative SDS-PAGE. The purified GST-FtsZ fusion protein was used to immunize rabbit to raise polyclonal antisera. Western analysis results showed that the expression level of FtsZ was tissue-specific: young flower> young leaf> young root> old leaf> stem. Accumulation of FtsZ was reduced by dark treatments. At the same time, the chloroplast turned to pale plastids with reduced volume and increased chloroplast number in cells. This showed that expression of FtSZ might not be regulated by light directly, and the expression of FtsZ played an important role in the division of plastid (chloroplast) and the number of plastid (chloroplast) in cells.
