Peach (prunus., Persica (l). Bastch) Germplasm Resources, Identification And Genetic Relationships By Rapd Analysis

Peach (prunus persica L Bastch) ,originating from the western of china ,has developed many species and categories in the long period of natural selecting and artificial domestication. About the identification,taxonomy and evoluation of peach, lots of work have been done in morphology,pollen and isoenzyme with some performance. However, there are still some divergence in result and limitiation in technology.RAPD has been used widely in species identification ,geneomy diversification examing and system analysing ,proved its value and effection.this reseach promulgated plentiful divercification of DNA in peach through RAPD , inquired into the taxonomy and evoluation of peach ,and attempted the feasiablity and method of RAPD in peach identification,to provide some basis to reseach and manufacture .The main results as follow:(1)48 peach species and categories were studied by selected 13 primers and 47 divercification seats. All the metierial were taxonomied into 5 typies at 130.0 distance:Prunus Kansuensis Hehd; Prunus Davidiana Carr; northern peach ; southern peach ; American peach. Ancient peach species were taxonomied in ecotype as the fires grade and in the character of fruit as the second grade,mordern species were done rightly in the character of fruit .Despite the latter ,peach may be taxonomied into the following typies: the middle of china peach, the northern peach,xinjiang peach, american peach. In each ecotype,by the character of fruit,peach species may be taxonomied into stone pulp peach,sweet peach(honey peach),flat peach,bare peach,yellow peach,and so on.(2)Ancient species and categories have obvious regional characteristic ,and mordern species heredity background is similar and the regional characteristic is not so abvious.American peach ,as a whole, may exist one ancient evoluation way. Xinjiang peach is similar with the northern peach,both of them may originate from stone pulp peach . Prunus Kansuensis Hehd and Prunus Davidiana Carr are most ancient in all the metierial .the northern peach, the southern peach and american peach are similar in evoluation status, which may rightly originate from Prunus Kansuensis Hehd and Prunus Davidiana Carr .(3)All the metierials were indentified with 6 primers and 15 fragment;as a result,except for BaiLu and YuHuaLu peach,all the metierials were effectively seperated.(4)The effect of RNA and protien to DNA examining was analysied. For the fact of DNA rough collecting ,DNA density was examined only by DNA electrophoresis result. The improved PCR programme includes 5 steps: advance denature, advance amplify,primary amplify,secondary amplify, complete amplify; according the differet amplifying aid in different step, each parameter is different slightly.
