In the process of culturing Cordycepts Militars, we recently found that their mycelium contains the protein that possesses the characteristics of fibrinolytic activity-Cordycepts Militars Fibrinolytic enzyme.After the cultured Cordycepts Militars was homogenized by arenaceous quartz grinding in 10mMPB (pH 8.04), toe coarse enzyme solution may be obtained by 70% (NH4)2SO4 precipitation and centrifuge.This component possesses the fibrinolytic activity that degrades fibrin and forms a transparent band by degrading fibrin on a porcine fibrin plate. Its optimal reaction temperature is 40℃ and optimal pH is 7.4. Certain concentration of Ca2+, Mg2+, Zn2+, Cu2+ ,Mn2+ and Hg2+ has a different inhibitory effect to Cordycepts Militars Fibrinolytic enzyme activity.Preliminary testing to the sample after CM anion exchange chromatography and Sephacryl S-200 gel filtration ,showed its molecular weight is approximately 31800(31.8KD).No fibrinolytic activity was detected in the sorophore of Cordycepts Militars ; but was detected in the mycelia and its culture medium. The peak fibrinolytic enzyme production lies in the mycelia proliferation phase, light will inhibit the fibrinolytic enzyme production. When cultured in solution, large amount of fibrinolytic enzyme exocytosis will occur. High fibrinolytic activity win be maintained under low temperature and natural dry condition. Little residual enzyme activity remains after title storage in 4℃ or 35℃ condition.Research showed that various conditions will affect the growth of Cordycepts Militars. Mycelia will grow rapidly in pure rice culture medium More Mycelia growth days needed in the rice medium that contains peptide. The sorophore growth showed to be thin and short under the fully lighted environment. Scattered light will grow a longer and bigger sorophore, and a single branch is weightier.The sorophore showed a distinctive growth pattern in the rice medium with peptide comparing to the pure rice medium. With good ventilation, the grown sorophore morphology is better.When culturing sorophore using mori, we can obtain the complete product similar to the natural Cordycepts Militars. Its conversion rate is approximately 20-30%.
