Thrombus-disease damage people's health seriously, its morbidity is high and rising year by year. So developing an effective, safety and low side-effect medicine is a hot spot problem. The Chinese herb has been used to treat thrombus-disease for thousands years, lots of Chinese herbs are used to treat thrombus-disease, especially compounded medication. Studying Chinese herb through biological chemical method proved to be prospective.Proteins, extracted from 45 Chinese herbs through 80% ammonium, were screened by fibrillarin-plate to define their fibrinolytic activities in vitro. The result showed that 22 species could decompose fibrillarin in vitro and go into effect in different time. Semen Myristicae, Trichosanthes Kirilowii Maxim, Pollen Typhae have notable effects, the diameters of dissolving-circle after 18 hours are 12mm, 16mm, 11mm. Considering both extracting quantity and specific activity, Leonurus japonicus, Myristica fragrans, Rubia cordifolia, Trichosanthes Kirilowii, Panax Pseudo-ginseng, Achyranthes bidentata, Typha angustata, Platycodon grandiflorus are suitable for purifying.The proteins of Achyranthes bidentata and Rubia cordifolia deposited by 80% ammonium were purified through anion exchange, testing the activity of every component, Achyranthes bidentata 40%-1, 40%-5,60%-1, 60%-3 and Rubia cordifolia 40%-1, 60%-1, 60%-2, 60%-3, 60%-4, 80%-2 have high specific activity. Achyranthes bidentata 40%-5 component has high specific activity(297.72 U/mg), molecular weight is about 24KD, and it is a monomer, PAGE and SDS-PAGE showed same single band, has high value for further study. Analyzing the PAGE result of purified component of Rubia cordifolia, single band with activity was not obtained.
