Calli were induced from immature embryo of maize (Zea mays. L.) inbred lines 18-599W, 18-599R, R15, R08, R09, S37, A318, Zong 31, C501, XianzaoH.The endogenous hormones of immature embryo and calli were measured with ELISA. The interrelations between endogenous hormones and the frequency of Callus type II were studied.Through analyzing the endogenous hormones content in different immature embryo and calli, the content and mixture ratio of endogenous hormones were one of the key factors that can affect callus induction frequency. In different immature embryo, the content of endogenous ABA, IAA and the rate of endogenous ABA/GA3 show great positive relativity to the frequency of callus type II. In callus type II, the content of endogenous ABA show positive relativity to the frequency of callus type callus type II. And the endogenous IAA and GA3 show negative relativity. The content of endogenous IAA, DHZR, ZR, EPA, GA4 in callus type II is higher than them in callus type I. To the contrary, endogenous ABA and GA3 is higher in callus type I.The result of analyzing the content of endogenous hormones during the period of embryogenic culture showed that endogenous ABA and IAA were the key factors to inducing the callus type II. During the period of embryogenic culture, the content of endogenous ABA, IAA, ABA/IAA, ABA/GA3 increased. Following the callus type II appearing, their contents attained the highest. Then their contents began to descend rapidly. To be different, the content of endogenous GA3 attained the lowest when the callus type II appeared.
