Compared with conventional inbred lines, the remarkable advantage of the doubled haploids lines is shorter breeding cycle and higher breeding efficiency when it’s used in practice. This article gives the method of solving these difficulties, specifically which is the lack of the technology in selecting haploid and low efficiency of doubling haploid in application of haploid breeding technology. In order to observe the purple marker in diploid immature embryo stage, we controlled the culture condition in terms of the color of illuminant, the intensity of light and the temperature. Then we found the best condition for obseving purple marker. We regulated the treating time and the treating concentration to select the optimal combination. What we have done made a contribution to haploid doubled. The results showed that 2000 Lux of red light will shorten time of appearance of the purple marker. Temperature has no impact on it. The more mature the embryo is, the more quickly the marker appearance. Treating the haploid embryo with 0.04% colchicine for 24 hours is the best combination, either for vitality or for the high efficiency of doubling.
