A set of monosomic addition lines of Beta corolliflora were the progenies from the hybridization of B. vulgaris L. and B. corolliflora Zoss. in sugar beet. One of these monosomic addition lines, designated as M14, which constituted of the normal 18 B. vulgaris chromosomes with the NO.9 chromosome of B. corolliflora, was found having a chromosome transmission frequency 97%. The reason that the transmission frequency in M14 was so high was interpreted as a possible occurrence of apomixis in M14. In order to isolate and identify the apomixis gene(s) of M14, the specific expression clones had been obtained by the method of SSH (suppression subtractive hybridization) and later had all been sequenced to become 211 ESTs. By using the function of DNAMAN software, 183 ESTs of 211 can not be translated completely into amino acids, the other way round 28 ESTs remained. In addition, 4 valued ESTs (Me341, Me263, Me219, Me206) of the 28 ESTs were selected as candidated gene in order to get complete gene. The RT-PCR experiment approved that they are specific expressed in M14. In the end, the complete Me341 gene of the 4 candidated genes was cloned by method of RACE (rapid amplification of cDNA end) .Through bioinformatic analyzing; its initial codon was located in the 372bp of the complete gene, encoding 230 amino acids. The BLAST result reveals that Me341 gene share 88% similarity with C class floral identity transcription factor AGAMOUS gene in Spinacia oleracea. Much work needs to be done about the structure and function of the gene in the future.
