In order to study the apomixis of Iris domestica,Iris domestica was crossed with three different colours of I.dichotoma.We observed the reciprocal F1 hybrids of three different colours of Iris dichotoma and I.domestica.The offsprings derived from I.domestica ×I.dichotoma were self-pollination,and the morphological characters were observed.Authenticity of 13 individual F1 hybrid of six interspecific crosses between three different colours of I.dichotoma and I.domestica were analyzed using single nucleotide polymorphism(SNP)markers.The pollen rube behaviors of I.domestica × I.dichotma and self-pollination of I.domestica was observed by pollen tube fluorescence observation.Paraffin sectioning was used to observe the double fertilization process of I.domestica x I.dichotma and self-pollination of I.domestica.The progeny of I.domestica as the pod parent and I.dichotoma of different flower colours as pollen parent were similar to the female parent,wheras the reciprocal crosses were different form both of their parents.The traits of the self-pollination progenies showed that they are similar to I.domestica and there was no traits separation.All of the F1 hybrids were successfully identified by SNP markers analyzation.When three different colours of I.dichotoma were used as female,all of the offsprings were genuine hybrids.On the contrary,the reciprocal crosses were false hybrids.SNP markers were successfully applied to identification of F1 hybrids of six interspecific crosses between I.dichotoma and I.domestica for the first time,which would provide a reference means for authenticity identification in Iris cross breeding in the future.I.domestica was identified apomixis successfully.The pollen rube behaviors of inter-specific pollination between I.domestica and I.dichotoma showed that pollen tube stagmated on the stigma of female parent or grew slowly.Self-pollination ofdomestica and inter-specific pollination between I.domestica and I.dichotoma embryology development situation showed that I.domestica which self-pollination completed the double fertilization process in 3 days after pollination,but we did not find the double fertilization process in inter-specific pollination between I.domestica and I.dichotoma.
