Diamondback moth (Plutella xylostella L.)(Lepidoptera) is the main pest of crucifer throughout the world. At present, the resistance management of diamondback moth has been the hard and serious problem all of the world. Diamondback moth has developed resistance to almost all kinds of insecticide. And the mechanisms of insecticide resistance are very complex. They are mainly correlated with the penetration of insecticide, the enzyme for metabolism and the insecticide targets. The different insecticides have different mechanism, so the mechanisms of insecticide resistance of diamondback moth are also different. But at last the mechanisms of insecticide resistance can resulted from the interaction of some genes which are involved in the insecticide resistance. So we believe that it is necessary for us to analyze the difference between susceptible strain and resistance strain of diamondback moth in genome level.In this study, we use susceptible strain, dimehypo resistance strain, cartap resistance strain and deltemethrin resistance strain of diamondback moth as materials, through representational difference analysis to analyze the difference of susceptible strain and resistance strain in genome level, and we also analysis the sequences we have got. Followed are our main results:1 In the research of deltmethrin resistance strain of diamondback moth, we got some differential bands which size varied from 200bp to 300bp.In the analysis of dimehypo resistance strain and cartap resistance strain, we got differential bands of 250bp.2 We purified these differential bands and cloned them. Every resistance strain ,we selected 20 positive clones at random and sequenced them. After we got these sequences, we made BLAST analysis on NCBI.3 In the analysis of deltmethrin resistance strain of diamondback moth, we got one sequence Di8 which is correlated with cytochrome P450; In the analysis of dimehypo resistance strain, we got three sequences:S109, S106 and S104. S106 is correlated with potassium voltage-gated channel; S109 has some homology with Cl channel; S104 has high homology with one cDNA sequence in Drosophila melanogaster adulttestis; But in the analysis of cartap resistance strain, we did not get sequences with high homology.4. As to Di8 sequence, it is correlated with cytochrome P450. It has been proved that cytochrome P450 determined the insecticide resistance; SI06 sequence show homology to potassium channel sequence;S109 has something with chloride channel. They all have important roles in neural system of diamondback moth. The NTX insecticides all have the neural system as their target. The changes in targets may decrease the effect of NTX insecticide; SI04 sequence has relationship with the propagation of diamondback moth. We believe that the unstable of insecticide resistance is coincident with the decreasing ability of propagation of diamondback moth.
